Refugium causing a mini cycle?

I'm in the process of helping my brother build a 10g fuge for his 29g BC. Last night we got everything siliconed in place and will be doing a leak test tomorrow after it's had 48hrs to dry. What I would like to know is if it will cause his tank to go into a mini cycle from adding new sand and LR to the existing setup? He's taking the sand and LR from a 3g pico tank that has been running for a few months that is stocked with only a couple hermits and snails. We're algo going to put some macro algae/chaeto in it.

well it should not since there is nothing in the sand and the rock is already cycled. but there is always a chance of it happing but very unlikely

I doubt it. He'll probably be okay.
